Increase of RNA Polymerase Activity in Pea Buds Treated with Gibberellic Acid A_3

• To determine the effects of gibberellic acid A_3 (GA_3) on the transcriptional machinery of the pea (Pisum sativum, var. Alaska), DNA-dependent RNA polymerase (nucleosidetriphosphate:RNA nucleotidyl transferase, EC. 2.7.7.6) activities from GA_3-treated tissue and control tissue were measured by a method involving partial purification by sequential elution on a DEAE-Sephadex A-25 column. We found that GA_3 enhances both RNA polymerase I and II activities but produces no detectable change in the properties of RNA polymerase II. Our data suggest that GA_3-dependent activiation of RNA synthesis is caused by an increase in RNA increase activity together wit~ enhancement of the template activity of chromatin (Tomi et al. 1983).
