Optimal Culture Conditions for Production of the Macromolecular Peptide Antibiotics AN-1,AN-3 and AN-7

The optimum cultural conditions giving maximum productivity of three kinds of macromolecular peptide anribiotics(AN-1,AN-3 and AN-7)were investigated. Calcium ion was essential for the biosynthesis and excretion of each antibiotic, and the antibacterial activity of these antibiotics was found only in the media, not within cells. Glucose was stimulative at lower concentrations for antibiotic production but markedly inhibitory when supplied at more than 40 mg/ml. AN-1 amounting to 280 units/ml(241 μg/ml)was produced by Streptomyces albus AJ9003 in a medium containing glucose, NH_4NO_3,Polypepton, dried yeast, yeast extract, CaCl_2 and ZnSO_4. AN-3,at 120 units/ml(1,104 μg/ml), was accumulated by Streptomyces albulus AJ9422 in a medium containing glucose, NH_4NO_3,Polypepton, dried yeast, meat extract, CaCl_2 and MnSO_4. AN-7,at 200 units/ml(63.5 μg/ml)was excreted by Streptomyces griseoincarnatus AJ9424 in a medium containing glucose, (NH_4)_2SO_4,Polypepton, dried yeast, meat extract, CaCl_2,MnSO_4 and MgSO_4.
