Biosynthetic Correlation between Streptomycin and Mucopeptide in Utilization of D-Glucosamine as a Common Precursor

When D-[1-^<14>C]-glucosamine was added to a mycelium-suspended culture of Streptomyces griseus, the radioactive carbon was incorporated into streptomycin as well as into mucopeptide. when the mucopeptide synthesis was suppressed with enduracidin, vancomycin or bacitracin, the incorporation of radioactivity into streptomycin was enhanced, suggesting that interruption of phospholipid cycle in mucopeptide synthesis was an effective trigger for shunting utilization of D-glucosamine towards streptomycin production. The effect of suppression of mucopeptide synthesis varied with the culture age of the mycelium, with most remarkable effect in the late exponential phase.The time of addition of radioactive D-glucosamine influenced the incorporation of radioactivity into the three molecular subunits of streptomycin. On addition at the start of culture, as much as 81% of the radioactivity was located in N-methyl-L-glucosamine, while on addition after 12 hr, 72% of the radioactivity was found in streptose. The streptidine moiety took up much less radioactivity than the other two subunits.
