DEFECTIVE ESTROGEN RECEPTOR IN UTERI OF RATS TREATED NEONATALLY WITH A HIGH DOSE OF ESTROGEN

雌S-D系ラットに出生直後より連日10日間estradiol-17β,10μgの皮下投与を行ったestrogen処置ラットでは,膣細胞診および組織学的検討から,成熟期に至るも持続的diestrusの状態を呈するのが認められた.放射性estrogenを投与すると,estrogen処置を受けた成熟ラット子宮ではestrogen の取込み量は無処置ラットに比し有意に低下したが,肝臓,横隔膜および血中放射能については有意差は認められなかった.蔗糖密度勾配遠心法では,無処置群の未熟および成熟ラット子宮のcytosol中には8S estrogen受容体が認められたが,estrogen処置群の成熟ラットでは屠殺時期や卵巣摘出の有無に関係なく8S estrogen受容体は検出されなかった.以上の結果から,出生直後の大量estrogen投与は,長期的効果として標的臓器である子宮のestrogen受容体欠損を惹起することが示唆された.この点に関し,子宮estrogen 受容体の発生時期と子宮内膜上皮の分化時期との関連性につき考察を加えた.
