INHIBITION OF TUMOR ANGIOGENESIS ACTIVITY IN C_3H MOUSE MAMMARY TUMOR BY MEDROXYPROGESTERONE ACETATE

C_3Hマウス乳腺腫瘍は, その増殖においてホルモン依存性がないと考えられている。事実, この移殖乳腺腫瘍の増殖やこの細胞のコロニー形成に, medroxyprogesterone acetate (MPA) は, 影響しない。この乳腺腫瘍組織およびこの細胞のコロニーのtumor angiogenesis factor (TAF) 活性は, それぞれ393.5%と46.8%で, fibroblast growth factor (FGF) 活性は, それぞれ49.6%と11.3%であった。乳腺腫瘍を移殖したC_3Hマウスに0.67 mgのMPAもしくはcortisolを毎日7日間皮下注射すると, この乳腺腫瘍組織のTAF活性は, それぞれ100.0%と325.5%で, FGF活性は, 23.7%と54.7%になった。また, 10^<-6>MのMPAもしくはcortisolと2週間培養して生じたコロニーのTAF活性は, それぞれ9.8%と44.3%で, FGF活性は, 2.7%と11.3%になった。したがって, MPAは, cortisolのもつようなgluticorticoid作用とは別に, またこの乳腺腫瘍細胞の増殖抑制とは無関係に, この腫瘍細胞のもつTAF, FGF活性を抑制し, 周囲の間質組織からの新生血管の生長を抑制すると考えられた。
