NUCLEAR TRANSLOCATION OF PROGESTERONE RECEPTOR-PROGESTOGEN COMPLEX IN VITRO

天然と合成のプロゲストーゲンの効果発現機構での違いを知るために,ウサギ子宮で各種プロゲストーゲンやその代謝物とプロゲステロンレセプター(PR)複合体の核内移行能について検討した.in vivoでのステロイド代謝による影響を避けるためin vivoの系を考案した.即ち子宮サイトソールとプロゲストーゲン及びその代謝物とを結合させ,この結合体と核分画への結合をin vivoの系でおこない,核におけるプロゲストーゲン及びその代謝物により取り込まれたPRを^3H-プロゲステロンによるexchange法にて測定した.プロゲステロン,デトロゲステロン,ノルエシンドロンは^3H-プロゲステロンとサイトソールPRとの結合の^3H-プロゲステロンに対してほぼ等しくexchangeされた.しかし,核に取り込まれたPR量をexchange法で測定した場合プロゲステロンがもっとも多くPRを核に取り込んだ.したがってプロゲステロンはそのレセプターをもっとも細胞質より核に移行させうると考えられ,ノルエシンドロンやデトロゲステロンの強力な生物学的活性の発現機構は他の面より考える必要がある.
