Many reports have suggested that platelets are related to the onset of bronchial asthma. These results stimulated us to clarify the role of platelets in bronchial asthma. The number of platelets, free calcium concentration, ATP content, and platelet aggregation in platelets taken from anaphylactic guinea pigs with or without asthma attack were measured with or without pretreatment of the antiallergic agent, ketotifen. Asthma induced by ovalbumin aerosol inhalation in immunized guinea pigs was effectively inhibited by the pretreatment of ketotifen. The number of platelets during asthma increased significantly, and ketotifen inhibited the increase. PAF and ionomycin inhibited both aggregation and free calcium concentration responses, although ADP, collagen and thrombin did not change the platelet responses. TPA showed a decreasing tendency in platelet aggregation and free calcium concentration. From these data, it was estimated that 1) antigen stimulates the mast cells and platelets, and that PAF is released from the cells, 2) PAF causes inflammation and contracts the bronchial muscle, and stimulates the platelets, and PF-4 and PAF are excreted, 3) these agents contract the bronchial smooth muscle, 4) PAF and SRS-A contract the bronchus and cause inflammation of the bronchus, 5) platelet disorders mentioned above initiate and accelerate the onset of bronchial asthma.
