We investigated responses to vasocontractile agents in the pulmonary artery and aorta of 1.5,3 and 6 month-old stroke-prone spontaneously hypertensive rats (SHRSP) and Wistar Kyoto rats (WKY). Results regarding the pulmonary artery showed that contractions by noradrenaline (NADR), phenylephrine (PHE) and methoxamine (MET) in the 1.5,and 3 month-old SHRSP did not differ from those in the age-matched WKY, although those in the 6 month-old SHRSP were greater than those in the age-matched WKY. The contractions by NADR were increased by N^w-nitro-L-arginine (LNAME) and propranolol and were more remarkable in WKY than in SHRSP. Although the contractions by serotonin (5-HT) did not differ between SHRSP and WKY at the 1.5 and 3 months of age, those in the 6 month-old SHRSP were greater than those in the age-matched WKY. The contractions by angiotensin II (ANG) were greater in SHRSP than in WKY at any age. Vasopressin (VAS) produced the small contractions only in the 6 month-old rats. The contractions by prostaglandin F_<2α> (PGF) in the 1.5 and 3 month-old SHRSP were greater than those in the age-matched WKY, but those in the 6 month-old SHRSP did not differ from those in the age-matched WKY. The contractions by KCl (K^+) did not differ between SHRSP and WKY at any age. Results regarding the aorta showed that contractions by NADR and by 5-HT were greater in SHRSP than in WKY at any age. Although the contractions by ANG in the 1.5 and 3 month-old SHRSP were greater than those of the age-matched WKY, those in the 6 month-old SHRSP did not differ from those of the age-matched WKY. The contractions by VAS in the 3 month-old SHPSP were greater than those in the age-matched WKY, those in the 6 month-old SHRSP were smaller than those in the age-matched WKY. The contractions by PGF did not differ between either strain at any age. The contractions by K^+ were greater in SHRSP than in WKY at any age. These results suggest the following (1). The vascular response to an agent is different between SHRSP and WKY and their difference in response is also different among various agents (2). NADR releases more EDRF in the pulmonary artery and the aorta, being larger in WKY than in SHRSP (3). The Ca^<2+> entry through depolarization in the aorta of SHRSP is accelerated compared with that in WKY from the results of K^+-induced contraction.
