The present study carried out on the responses to vasocontractile agents in the renal arteries and the aortas from the stroke-prone SHR (SHRSP) at the 1.5 months (prehypertensive stage), 3 months (stage of developmental hypertension) and 6 months (stage of duration of hypertension) of age and compared with those of the age-matched Wistar Kyoto rats (WKY). Results in renal artery : The contractile responses to the alpha-receptor stimulating agents, noradrenaline (NADR), phenylephrine (PHE), methoxamine (MET) were not different between SHRSP and WKY at 1.5 months of age. The contractile responses to the lower concentrations of those agents at the 3 month-old SHRSP were greater than those of the age-matched WKY, although those in the higher concentrations were not different between the both strain. The same trend was found at the 6 month-old SHRSP and WKY, although the responses to MET and PHE in the extreamely high concentrations were smaller in SHRSP than in WKY. The contractile responses to serotonin (5-HT) were not different between SHRSP and WKY at any age tested. Although the contractile responses to vasopressin (VAS) were not different between SHRSP and WKY at 1.5 and 3 months of age, those at 6 months of age were smaller in SHRSP than in WKY. The contractile responses to angiotensin II and K^+ were greater in SHRSP than in WKY at any age tested. The pD_2 values of NADR, PHE, MET, 5-HT, ANG and VAS were not different between SHRSP and WKY at any age tested, and also were not changed with the age in both SHRSP and WKY. Results in aorta : The contractile responses to NADR, PHE and MET were slightly greater in SHRSP than in WKY at any age, although the difference was not significant. The same trend was found in the contractile resposes to 5-HT. In the case of VAS and and ANG, the contractile responses at the 1.5 and 3 months of age were greater in SHRSP than in WKY, but those at the 6 months of age were greater in WKY than in SHRSP. The contractile responses to K^+ were greater in SHRSP than in WKY at any age. The pD_2 values of NADR, PHE, MET, 5-HT, ANG and VAS were not different between SHRSP and WKY at any age and also were not changed with the age. These results sugges that the increased voltage-dependent Ca^<2+> influx is a important factor for the development and the maintenance hypertension of SHRSP and the participation of receptor-mediated Ca^<2+> influx for the hypertension of SHRSP is different from the kind of agents and from the kind of blood vessels, and the quality of receptor is not changed with the age and is not affected by the modification of blood pressure.
