This paper demonstrates the concept of a diagnosis chip for infectious diseases caused by viruses, and especially the one-chip integration of DNA purification, trapping, and detection. We attempted to construct a DNA purification tool (DNA pre-treatment device) comprising alumina (Al2O3)-coated micro-pillars and hydrogel valves, whose function varied with the pH of washing solutions. It was found that DNA was immobilized on the micro-pillars in acidic solution, eluted in alkaline solution, and reversed by controlling the pH-dependent hydrogel valves. As a result, one-chip integration for infectious disease diagnosis combined with a DNA cleaning chip was achieved.
