3′-Deoxykanamycin has been totally synthesized from 6-<I>O</I>-(2-<I>O</I>-benzyl-3-deoxy-3-ethoxycarbonylamino-4,6-<I>O</I>-isopropylidene-α-D-glucopyranosyl)-<I>N</I>,<I>N</I>′-diethoxycarbonyl-2-deoxystreptamiine and 6-azido-2,4-di-<I>O</I>-benzyl-3,6-dideoxy-α-D<I>ribo</I>–hexopyranosyl chloride. It has been found that the 3′-deoxykanamycin has strong antibacterial activity not only against common bacteria but also against kanamycin-resistant bacteria.
