A hot-water extract was prepared from delipidated <I>Mycobacterium bovis</I>, strain ECG. Chemical analyses revealed that this extract consisted of neutral sugars, peptides and nucleic acids which were presumably derived from the cytoplasmic fraction. When this extract was in jected into guinea pigs in combination with Freunds incomplete ad juvant plus egg white albumin as antigen, an increase of antibody titer was observed and delayed hypersensitivity to egg white albumin as evidenced by positive skin and corneal reactions was induced. Thus the hot-water extract was found to be ad juvant-active. However it seems unlikely that its ad juvant activity was due to the concomitant presence of nucleic acids, since the ad juvant activity was retained even after removal of nucleic acids.<BR>No undesirable side effects were observed in mice which had received intraperitoneal in jections for 20 consecutive days of 0.25mg of this extract.<BR>An antitumor activity was observed when this extract was in jected intraperitoneally after an intraperitoneal inoculation into mice of Sarcoma 180 cells or Ehrlich carcinoma cells. A combined effect on Sarcoma 180 and Ehrlich carcinoma was found when this extract was in jected in combination with Mitomycin C or cyclophosphamide, respectively. Intraperitoneal injections of this extract prolonged the survival time of C3H/He mice which had received an intraperitoneal inoculation of 5×10<SUP>3</SUP> cells of a syngeneic ascites hepatoma, MH134.
