The growth of L. bifidus A-3 was observed using homogenated faeces of infant by tenfold dilution as the medium. On the media prepared from faeces of breast-fed and mixed-fed infants, the strain showed distinct growth as measured by acid production. In contrast, it exhibited only scant or undetectable growth on the media prepared from faeces of bottle-fed infants. However, it showed marked growth if the media were supplemented with lactose. Large amounts of oligo- and polysaccharides were found in the media prepared from faeces of breast-fed infants. In addition, small amounts of galactose and fucose, and negligible or no lactose and glucose were also detected. After incubation of the strain, a marked decrease of oligo- and polysaccharides and an increase of fucose and galactose were observed. Only small amounts of polysaccharides and traces of galactose and fucose were detected in the media prepared from faeces of bottle-fed infants. After incubation of the strain, their change in quantity was undetectable. The amount of sugars in the media prepared from faeces of mixed-fed infants were varied and usually low. However, the presence of oligo- and polysaccharides and their decrease after incubation of the strain were clearly observed. From the results obtained, it would seem that the amounts of sugars available for L. bifidus in the intestinal content would affect the difference of intestinal flora observed between breast-fed and bottle-fed infants.
