The process of adrenocortical regeneration after bilateral enucleation was studied in rats by examining the rate of incorporation of labeled nucleic acid precursors, together with DNA and RNA determinations. It was concluded that adrenal enucleation induced rapid cellular proliferation which was almost completed by the 20th day after operation. Quantitative changes in adrenocortical components, which are considered to be related to corticosterone biosynthesis, were also investigated during the course of adrenocortical regeneration provoked by bilateral enucleation, along with the responsiveness of some of the components to ACTH administration. Levels of adrenal ascorbic acid, cholesterol, cholesterol sidechain-cleaving activity, cytochromes and proteins of subcellular particulate fractions as well as of plasma corticosterone, showed decreasing tendency after bilateral enucleation. Rates of the initial decrease and later recovery were varied between components. On the 20th day after operation when cellular proliferation was considered to be almost completed, these cellular components, except for nucleic acids, microsomal protein and plasma corticosterone, were still at lower levels than in intact controls. It was suggested, therefore, that a lag time would be necessary for the recovery of function after completion of cellular proliferation.Physiological significance of the amounts of cellular components was discussed in relation to the maintenance of plasma corticosterone level.
