Identification and Inhibitory Effect of Fatty Acids from Hyperici Herba on Bacterial Sialidases

• An aqueous acetone extract of Hyperici Herba had an inhibitory effect on the bacterial sialidase from Clostridium perfringens (Cl-S). An active principle was purified from the extract by silica gel column chromatography. The active principle was a fatty acid mixture consisting of palmitic acid, oleic acid and linoleic acid as determined by gas-liquid chromatography (GLC) and combined GLC-mass spectrometry (GC-MS) of their methyl esters. The inhibitory activity of each of the three fatty acids on various sialidases was assayed. Oleic acid and linoleic acid were the inhibitors of Cl-S and also of the sialidase from Streptococcus spIID6646 (St-S). Palmitic acid inhibited only St-S. The three acids showed no effect on the sialidase from ArthroBacter ureafaciens (Ar-S). Oleic and linoleic acids competitively inhibited Cl-S. The concentrations of the sodium salts and methyl esters of these fatty acids in the assay mixture to give 50 % inhibition were higher than those of the respective free fatty acids.
