Objective : Cigarette smoking, as it relates to race and ethnic background, is associated with immunological alterations. The aim of this study was to clarify the effects of smoking on serum cytokine levels, as they differ among races and ethnic backgrounds of healthy men. Study Design : We measured serum levels of interleukin-4 (IL-4), IL-5, IL-6, IL-10, interferon-γ (IFN-γ) and granulocyte macrophage-colony stimulating factror (GM-CSF) using chemiluminescence enzyme-linked immunosorbent assay (CL-ELISA) in 372 healthy men, including Japanese, Caucasians and African Americans. Results : Cigarette smoking was associated with a significant decrease in serum IL-5 in the combined population, and in Caucasians, African Americans and Japanese analyzed separately. Among nonsmokers, Japanese men had significantly lower serum IL-5 than did either Caucasians or African Americans. The mean IL-6 in Japanese nonsmokers was significantly lower than that in African Americans. Japanese nonsmokers also had a significantly lower IL-10 than did Caucasians. These results were confirmed by multiple regression analysis. Conclusions : The differences in serum IL-5, IL-6 and IL-10 levels among races might be related to ethnic differences as they are revealed in allergic diseases such as asthma. Smoking and ethnic differences must be considered in population studies using serum cytokine measurements.
