Isolation and Identification of Self-Transfer Products Formed by Guinea Pig Liver β-Mannosidase(Organic,Chemical)

β-Mannosidase from guinea pig liver was found to catalyze the self-transfer reaction with aryl β-D-mannopyranosides. With p-chlorophenyl and p-nitrophenyl β-D-mannopyranosides, two corresponding aryl β-mannobiosides were isolated from both substrates as main transfer products by using a Toyopearl HW-40 column. The structures of the transfer products were determined by chemical and enzymatic hydrolysis, and mass spectrometry of the methylated alditol acetates. The results indicated that the enzyme transferred the β-mannose residue from the substrate to the hydroxyl group at the 2- or 6-position of another substrate molecule.
