Effects of basic fibroblast growth factor on high energy phosphate metabolism in the ischemic myocardium following ischemia/reperfusion-A preliminary report

Basic fibroblast growth factor (bFGF) has been shown in some studies to promote angiogenesis and myocardial salvage in experimentally induced acute myocardial infarction. We believe this is not solely caused by the angiogenic action of bFGF. To identify the possible action of bFGF, in the present study, we investigated the effect of bFGF on high-energy phosphate metabolism in the ischemic myocardium using ^<31>P-NMR. In Japanese white rabbits, the heart was perfused with Langendorff preparation. After basal perfusion for 10 minutes, global ischemia was induced for 20 minutes followed by reperfusion for 60 minutes. In the bFGF group, 100μg/0.1mL of bFGF were injected in the perfusate 1 minute prior to the induction of global ischemia. Then, we compared the bFGF group with the control group in terms of the high-energy phosphate metabolism. After 5 minutes of reperfusion, adenosine triphosphate (ATP) level was 59.4±5.5 (±SE)% in the bFGF group. Comparing this value with that of the control group (42.0±5.0%), ATP was maintained at a significantly higher level in the bFGF group. Our results suggested that bFGF maintains a high ATP level during ischemia, suppressing ischemia-induced myocardial injury.
