Morphological and Biochemical Studies on the Healing Process after Cryosurgery

In order to investigate the fundamental process of cryo-degeneration and regeneration, rabbit-back skin was frozen by the two-cycle freezing method and the cryo-lesions were examined histologically and biochemically. Histologically, necrobiosis and exfoliation of the epidermis and intensive edema of the dermis were observed at the center of the cryo-leison early in the first week. Tissue necrosis was seen late in the first week and primary regenerative changes of the epidermis were observed early in the second week as a mode of epidermal stretching from the margin. This stretching, however, sloughed off within a few days, probably due to inadequate granulation in the dermis, and the lesions were replaced by the epithelium of the secondary regeneration. Collagen synthesis of the dermis was analyzed by determining the radioactivity of ^<14>C-hydroxyproline after labeling the tissue with ^<14>C-proline. One week after the cryosurgery, the specific radioactivity of the bottom of the lesion was more than three times higher than the surface boundary. It gradually decreased thereafter, although it remained higher than normal.
