The Presence of An Estrogen Receptor in The Castrated Mouse Thymus

Using the techniques of sucrose density gradient ultracentrifugation and dextran-coated charcoal adsorption, the characterization of estrogen-binding proteins was attempted in cytosols from the thymus, spleen and mesenteric lymph node of castrated male and female mice of the C57BL strain. After incubation of the thymic cytosol of females with ^3H-estradiol-17β in the presence or in the absence of excess unlabeled steroids, specific estrogen-binding protein with a binding capacity of〜5×10^<-14> moles/mg protein and dissociation constant of〜9×10^<-9> M was found in the 4 S region. The cytosol of the male thymus also showed specific 4 S estrogen-binding with almost the same values of binding capacity and dissociation constant as those of female cytosol. No specific binding, however, was found in the cytosols of the spleen and mesenteric lymph node of either sex. These results suggest that castrated male and female mice contain a specific estrogen receptor in their thymi.
