Uptake and Binding of ^3H-testosterone by Lymphoid Tissues of Castrated Mice

The in vivo uptake of ^3H-testosterone by the lymphoid tissues (bone marrow, thymus, mesenteric lymph node and spleen) of castrated male and female C57BL mice was studied following an i. v. injection of the ^3H-labelled hormone. All the lymphoid tissues in females took up more radioactivity than those in males. In addition, radioactivity uptake and retention in the tissues of both sexes was relatively higher than those in the serum. The binding of ^3H-testosterone by the cytosols of lymphoid tissues was examined in vitro by Sephadex G-100 column chromatography. Cytosol from all tissues contained the testosterone-binding component(-s), probably protein(-s). These results suggest that there is relatively high uptake and retention of testosterone in the lymphoid tissues in which it is bound to cytoplasmic binding component(-s).
