Cycloprodigiosin Uncouples H^+-Pyrophosphatase of Plant Vacuolar Membranes in the Presence of Chloride Ion

Cycloprodigiosin hydrochloride (cPrG-HCl) at 10 nM uncoupled both H^+-ATPase and H^+-translocating pyrophosphatase in vacuolar membrane vesicles of mung bean, but did not inhibit the substrate hydrolysis. cPrG-HCl destroyed the transmembrane pH gradient only in the presence of Cl^-. The observations indicate that cPrG-HCl functions as a H^+/Cl^- symporter. Vesicle acidification was restored by SO^<2->_4 even in the presence of cPrG-HCl.
