Purification and Characterization of Adenine Deaminase in Pseudomonas synxantha : Metabolism of nucleosides in bacteria (VIII)

A microorganism which grows on adenosine, as the soke carbon and nitrogen source, was isolated and identified as a variety of Pseudomonas synxantha. This strain assimilated various purines as the sole source of carbon an dnitrogen, and adenosine was the best nutrient source of all the purine compounds tested. In this strain, adenosine was degraded mainly via adenine. Adenine deaminase, which catalyzes the first step of degradation of adenine in this organism, was purified to an electrophoretically homogeneous state, and some properties were investigated. The molecular weight of the enzyme was around 37,000. The enzyme catalyzed the replacement by a hydroxyl group of the substituent in the 6-position of substituted purines such as adenine, 2-amino-6-chloropurine, 6-bromopurine, 6-chloropurine, 6-amino-2-hydroxypurine, 2,6-adenine, diaminopurine and 6-iodopurine, with decreasing rates in that order.
