Recovery of Intact Membrane Proteins from Adherent Animal Cells Grown in a Liquid-Liquid Interface

The detachment of adherent animal cells (A375; Human malignant melanoma cells) grown in plastic dishes by trypsin-treatment accompanied the proteolysis of membrane proteins. To avoid this proteolysis, A375 cells were grown in a liquid-liquid (culture medium and fluorocarbon fluid) interface and were easily harvested by pipetting with an intact form of membrane proteins. Though the shape of the cells grown at the liquid-liquid interface was slightly globular, subculturing from the interface to the same interface, to plastic dishes or to glass dishes was possible.
