Biotransformation of D-Xylulose to D-Xylose by Immobilized Enzyme Prepared from Streptomyces flavorirensis

Screening of Actionmycetes suggested that D-xylose isomerase activity is distributed widely in the genus Streptomyces. Streptomyces flavorirensis showed the strongest D-xylose isomerase activity of the tested strains, and its isomerization yield of D-xylose was 91.5%.For the purpose of enzymatic conversion of D-xylulose to D-xylose, properties of immobilized D-xylose isomerase prepared from S. flavorirensis were investigated. The enzyme activity was stable at high temperature(55℃) and high pH(9.0〜10.5), and was activatetd by Co<2+> and Mg<2+>. These properties were favorable for the continuous conversion of D-xylulose to D-xylose by immobilized D-xylose isomerase. In 8 days of operation at a space velocity of 0.25 h<-1>, 29.4 g of D-xylose was produced from 36.0 g of D-xylulose (yield 81.7%).
