Production, Purification and Some Properties of Extracellular Amylase of Schizophyllum commune

An extracellular saccharifying amylase from Schizophillum commune Fries was purified and its properties were studied. The productivity of the enzyme by S. commune was increased by addition of thiamine to the culture medium containing wheat bran. The enzyme was purified by ammonium sulfate precipitation, acid-clay treatment and Sephadex G-100 gel filtration to give a single band on a sodoium dodecyl sulfate polyacrylamide disc gel electrophoresis. The saccharifying amylase was found to be glucoamylase and to produce a large amount of glucose from raw rice starch and starchy materials.It was a molacular weight of 66,000 and is stable up to 55℃.
