Immobilization of an NADH Oxidizing Complex and its Use as a Regenerator of NAD

In order to obtain a stable and active NADH oxidizing complex, the activity of NADH oxidase was investigated in 39 strains of aerobic bacteria and yeast. The NADH oxidizing complex from Arthrobacter oxydans exhibited the highest activity among the aerobes investigated. The NADH oxidizing complex was fractionated by ultracentrifugation and immobilized with acrylic monomer on ^<60>Co-γ radiation. The activity shown by the immobilized enzyme complex was 20-50% of the initial activity. Enzymatic properties of immobilized NADH oxidizing complex were investigated in comparison with those of the free enzyme complex. In addition, the availability of the immobilized NADH oxidizing complex as a regenerator of NAD was shown using a simultaneously immobilized alcohol dehydrogenase-NADH oxidizing complex system.
