Comparison of Enzyme Activities in Oxalate Synthesis between Spinacia oleracea L.and Brassica campestris L.

Enzymatic conversion of glycolate to oxalate was studied in detached leaves of Spinacia oleracea L.(spinach), an oxalate accumulator plant and Brassica campestris L.(komatsuna), an oxalate non-accumulator plant by the application of 2-^<14>C-glycolate. The glycolate incorporated into leaves of the two species was rapidly metabolized. However, oxalate was not detected in the leaves of komatsuna. The in vitro experiment usin crude enzyme extract from leaves of the two species failed to reveal any appreciable differences in the enzymatic properties of glycolate oxidase between the two species. Therefore, the oxalate synthesis by the crude enzyme in spinach and komatsuna was studied using glutamate or alanine as amino-group donor. In both plants, oxalate synthesis in the presence of amino-group donor was reduced compared with that in the absence of amino-group donor. In addition, the extent of the decrease was larger in komatsuna than in spinach.
