ワンポットグリコシル化反応を基盤とする糖鎖迅速合成法の開発

本総説では、当研究室にて、研究してきたワンポットグリコシル化反応を利用した生物活性糖鎖の合成研究について述べる。ワンポットグリコシル化反応は、連続的な化学および、位置選択的なグリコシル化反応を同一反応容器内で行なうことにより、多種類の糖鎖合成ユニットより、オリゴ糖を一挙に合成する手法である。本手法を利用することにより、7つの独立な糖鎖ユニット利用した分岐7糖のワンポット合成が可能である。また、シアル酸を含む糖アミノ酸の合成も可能である。シアル酸含有糖鎖の合成では、シアル酸の5位のアセトアミド基をN-Troc基で置換したシアル酸糖供与体を利用した。また、ワンポットグリコシル化反応と並列液相自動合成装置を利用することにより、糖鎖のコンビナトリアルライブラリーの自動合成を達成した。さらに、得られた保護糖鎖の効率的な脱保護法として、固相脱保護法についても合わせて報告する。
