Biosynthesis of Mutant .BETA.-D-Galactosidases Containing Nonnatural Aromatic Amino Acids by Esherichia coli.

Mutant β-D-galactosidases containing nonnatural aromatic amino acids were produced by misincorporation during protein overproduction by <I>Esherichia coli</I>. The incorporation of 3-(1-pyrenyl)-L-alanine (pyrAla) was confirmed by fluorescence measurement of the produced enzyme, although the replacement frequency was low. Some of mutant enzymes showed improved thermostability.
