Limb allograft in rats immunosuppressed with Cyclosporin. Histologic grading.:Histologic Grading

As a skeletal tissue allograft model, limb allografts with or without Cyclosporin (CS) treatment using three inbred rat strains, Fischer (F344; RT11), Lewis (LEW; RT11) and Brown Norway (BN; RT 1n), were performed, and we graded the degree of rejection according to the categories by Fritz.In minor-mismatched allografts (LEW→F344), clinical and histologic examinations revealed no signs of rejection 16 weeks after operation with the use of CS, whereas, without CS treatment severe degeneration and necrosis of the grafts were shown by 16 weeks.In major-mismatched allografts (BN→F344), histologic examination revealed that rejection in CS-treated recipients did not appear as typical rejection, but only as a mild skin rejection, even though rejection appeared complete by gross visual observation of the skin 16 weeks after operation. While the grafts without CS treatment were degenerated and destructed by 6 weeks.Our results indicate the possibility of clinical application of skeletal tissue allografts with the use of CS as well as other organ transplantations.
