Root Differentiation and Paeoniflorin Production in Paeonia lactiflora Callus Cultures

Studies were conducted on effects of various plant growth regulators on root differentiation of paeonifiorin production by the callus derived from an immature anther of Paeonia lactiflora PALL, in static and liquid culture systems. Paeonifiorin and benzoic acid contents in EtOH extracts from the callus tissues were determined by high performance liquid chromatography. The highest paeonifiorin content (1.7% of dry weight) was obtained in the differentiated root derived from fairly wet and light brown callus line and cultured in the Linsmaier and Skoog basal medium containing 10^<-6> m 2,4-dichlorophenoxyacetic acid and 10^<-6> M kinetin, and then in the medium containing 10^<-7> m indole-3-acetic acid. These results showed that root cultures produced as much paeonifiorin as root of the intact plant.
