Determination of Arecoline and Arecaidine in Areca by Reversed-Phase, Ion-Pair High-Performance Liquid Chromatography

The chromatographic behaviors of arecoline and arecaidine were examined by reversed-phase, ion-pair high-performance liquid chromatography. The best separation of these alkaloids of areca was obtained by an isocratic chromatographic system using a mixture of H_2O-MeOH-SDS-phosphoric acid as the mobile phase at 70℃. Under the HPLC conditions selected, the two alkaloids gave large well-separated peaks and the detection limits were 0.01% at signal-to-noise ratio for each component. The analysis of 12 commercial areca showed that arecoline and arecaidine were contained at 0.09 to 0.68% (av. 0.31%) and 0.11 to 0.58% (av. 0.39%), respectively.
