Quantitative Analysis of Plant Hormones, Auxins, in Biotechnologically Cultured Products of Medicinal Plants

The biotechnologically cultured cells of Coptis plant and Lithospermum plant were analyzed for the residual levels of plant hormones, especially of auxins, 3-indoleacetic acid (IAA) and α-naphthylacetic acid (NAA). The procedures of clean-up and high-performance liquid chromatography (HPLC) using a reversed-phase column were studied. 0.1 n sodium hydroxide extracts of samples were successively partitioned between water and ether. Two solvent mixtures of water-acetonitrile-glacial acetic acid were used as the mobile phase for HPLC. A fluorescence monitor was applied whose detection limit of each auxin was 0.1 ppm in the samples. The recovery of spiked auxins at 5 ppm level by this method was almost quantitative (84.3-103%). When 1.9 ppm of NAA was used as the only auxin in the culture medium, 0.5 ppm of it remained in the fresh cultured Coptis cells. When IAA was used at the same level, it was not detected in the cells: it appears to have been comparatively used up easily during the culture.
