Standard Compounds for Quantitative Determination of Principles of Crude Drugs I : Paeoniflorin, a Major Principle of Peony Root

A procedure for the estimation of the purity of paeoniflorin, a major principle of peony (Paeonia albiflora PALLAS var. trichocarpa BUNGE) root, was established. The singleness of purified paeoniflorin was confirmed by thin layer chromatography, high performance liquid chromatography and ^<13>C-NMR spectrometry. Its purity was estimated by saponification of paeoniflorin and subsequent gas-chromatographic determination of benzoic acid produced. The rate of saponification is the same for the sample weight in a range of 10 to 40 mg. When 20 mg of the sample was subjected to the hydrolysis, benzoic acid was produced in 96.2±1.7% yield.
