In vivoならびにIn vitroで成熟させた異なる生殖周期に採取した犬の卵母細胞中のグルタチオン濃度(臨床繁殖学)

卵母細脳中のグルタチオン濃度は細胞質成熟のマーカーとして重要である.そこで,in vivoとin vitroで成熟させた犬卵母細胞中のグルタチオン濃度について比較検討した.In vivoで成熟させた卵母細胞は排卵72時間後に開腹し,卵管を環流して採取した.異なる性周期の卵巣を採取して得られた卵母細胞は,その直径が120μm未満と120μm以上の2群にわけ,それぞれ10%FBS,2.2mg/ml NaHCO_3, 2.0μs/ml estrogen, 0.5μ2/ml FSH,0.03IU/ml hCG,1%penicillin-streptomycinを添加したTCM-199培養液で,50μMβ-mercantoethanol添加あるいは無添加で72時間培養した.GSH濃度はDTNB-GSSG reductase recycling法で測定した.未成熟な犬卵母細腹中のGSH濃度は発情休止期,卵胞期,黄体期,それぞれの120μm未満と120μm以上の卵母細胞で,2.9と3.8,3.5と6.8,3.1と6.5pM/卵母細胞と有意に120μm以上の卵母細胞が高かった(p<0.05).In vivoで成然した卵母細腹中のGSH濃度(19.2pM/卵母細胞)はin vitroで成熟させた卵母細胞(β-mercaotoethanol無添加4.1-8.1pM/卵母細胞,添加5.7-13.2pM/卵母細胞)よりも有意な高値(p<0.05)であった.したがってβ-mercantoethanol添加はin vitroで培養した卵母細胞のGSH合成を増加させ,また卵胞期ならびに黄体期に採取した卵母細胞は発情休止期に採取した卵母細胞よりもin vitroの成熟に優れていると考えられた.
