The Delineation of Bursal Large Round Cells Increased by Neonatal Cyclophosphamide-Treatment of Chickens and the Role of the Cells for the B Cell Maturation

Large round cells were detected in the bursa of Fabricius of both normal chickens and chickens neonatally treated with cyclophosphamide. The cells were increased markedly, in striking contrast to the complete disappearance of lymphocytes, by the neonatal cyclophosphamide-treatment of the chickens. The cells were larger than lymphocytes in size, and were released from bursal tissues only the homogenization and trypsin-treatment. Rabbit antiserum against the bursal large round cells was prepared, and was absorbed with chicken red blood cells, serum, liver powder and lymphocytes. The antiserum thus abssorbed exhibited a high level of cytotoxicity against large round cells in the bursa but not in the spleen and thymus of 2-week-old chickens neonatally treated with cyclophosphosphamide. The absorbed antiserum was innocuous against lymphocytes from the bursa, spleen and thymus of 2-week-old normal chickens. Injection of the antiserum into newly hatched chickens markedly decreased the amount of the bursal antigen detected with this antiserum. The injection markedly decreased the number of spleen lymphocytes. The chickens injected with the antiserum had low capacity to produce plaque forming cells against sheep red blood cells and Salmonella pullorum. These results indicate that the large round cell in the bursa may participate in the intrabursal maturation of immature stem cells into immunocompetent B lymphocytes.
