Basic and clinical studies on newly developed oral cephalosporins such as Cefaclor, CGP-9000 and Cefadroxil are reported. With respect to absorption and excretion, CGP-9000 and Cefadroxil were not affected much by a meal, whereas absorption of Cefaclor was decreased by a meal as in the case of other oral drugs. By measuring MICs of each drug against E. coli isolated from the urine of out-patients, the antibacterial activity of Cefaclor was found to be most effective, and that of the other two were the same as that of Cephalexin. In addition, the results of clinical experience in out-patients with UTI : 50 cases given Cefaclor, 40 given CGP-9000 and 30 given Cefadroxil, showed that Cefaclor was the most promising drug.
