Vitellogenin (VTG) and estrogen receptor (ER) are estrogen responsive genes. Then these are using as biomarker for assessment of estrogenic activity of chemicals. However there are few investigations of VTG and ER gene expression pattern in early stage. In this study, we investigated VTG and ER time dependent gene expression in short exposure time using male Medaka. Medaka were exposed to estradiol-17 β (E_2) on 1, 4, 8, 12 hour, and measured hepatic VTG1, VTG2, ER gene expressions by reverse transcription polymerase chain reaction (RT-PCR). Consequently, these genes expression increased typically in 8 hour. In comparison between VTG1, VTG2 and ER, gene expression of VTG1 increased slower than VTG2 and ER genes. Thus male Medaka exposed to nonylphenol (NP), 4-(1-adamantyl)phenol (AdP), bisphenol A (BPA), BPA metabolite 4-methy 1-2, 4-bis(4-hydroxyphenyl) pent-1-ene (MBP), and 4, 4'-dihydroxy-α-methylstilbene (DHMS) for 8 hour and investigated hepatic VTG1, VTG2, ER gene expression and assessed estrogenic activity. As a result, AdP showed high estrogenic activity comparable with E_2. MBP and DHMS, which are metabolites of BPA, showed higher estrogenic activity than original compound. These results suggested that VTG2 and ER are useful biomarker for assessment estrogenic activity of chemicals, and there is a possibility that VTG1 and VTG2 are regulated by different gene expression systems.
