Bioimaging of protein secondary structure from Fourier transform infrared spectroscopy in experimental tumor tissue

We scrutinized the molecular changes in the necrotic core of an carcinoma tissue by infrared microscopy. We showed the significantly high accumulation of cholesterol linoleate in a necrotic tissue. The mechanism of this is hitherto unrecognized. We considered this could be caused by a membrane fusion by the biomembrane damage, membrane lipid shed and accumulated extracellulary. We revealed the ammounts of β-sheet significantly increased in striking contrast with the significantly decreased that α-helix in a necrotic area. We considered that the conversion was due to the lipid-auto-oxidation products like cholesterol oxide synthesized in a necrotic area, and the lipid-auto-oxidation products probably deformed protein secondary structure from an α-helix to a β-sheet.
