Diploid a or α sporogenous mater cells were frequently obtained in the sake yeast Kyokai no. 9 by heat treatment followed by random spore plating. Polyploid cells were constructed between the opposite mater cells by the mass mating technique, followed by the isolation of the zygotes using a micromanipulator.
