[SHORT REPORTS]Immunohistochemical Examination of Serotonin Fibers in the Developing Visual Cortex of Rats with Methylazoxymethanol-Induced Mictocephaly.-II

Treating pregnant rats with methylazoxymethanol acetate (MAM), an anti-mi-totic agent, on day 15 of gestation resulted in the formation of severe microcephaly in their offspring. This study immunohistochemically examined possible alteration in development of the 5HT fibers in the occipital cortex of microcephalic rats (MAM-rats) at age 7 and 9 days. the 5HT fibers rapidly increased in number and become more slender in the cerebral cortex of both the control and MAM-rats. 5HT fibers were irregularly and torthuosusky distributed in the MAM-rats. On the 7th postnatal day of the controls, a laminarly arranged, heavy aggregation of 5HT nerve terminals was observed, and a majority of the 5HT nerve terminals had become localizaed within layer IV of the cerebral cortex. In the MAM-rats of the some age, aggregation of the 5HT nerve terminlas was observed, and a majority of the 5HT nerve terminals had become localized within layer IV of the cerebral cortex, In the MAM-rats of the same age, aggregation of the 5HT nerve terminals was abnormally dense. These observations on the developmental abnormalities of 5HT fibers and terminals in the cerebral cortex within the first two postnatal weeks further support the morphological hypotheses that 5HT fibers play an important role in the development of neurons and layer meturation of the cerebral cortex and in the synaptogenesis of the thalamo-cortical input.
