Tumor Necrosis Factor (TNF) とInterleukin (IL)-1の直接注入による脳病変

マウス脳内にtumor necrosis factorα(TNFα)とinterleukin-1α(IL-1α)を直接注入して, 脳病変を経時的に検討した. 注入後約12時間で, 刺入部から数mm離れた小血管周囲に単核球を中心とした細胞漫潤がみられ, 24時間後も持続していた. またTNFα注入群の方がIL-1α注入群と比較して細胞浸潤が強かった. これらの病変は実験的アレルギー性脳脊髄炎(EAE)の初期と類似しており, TNFαとIL-1αによる血管透過性亢進が発症機序に関与している可能性が示唆された. EAEで使用する神経由来の抗原を用いなくとも, これらのcytokineを直接脳内に注入することで脳炎を惹起できる可能性があり, 多発性硬化症をはじめとした脱髄病変形成のメカニズムを考える上で重要である.
