PROTEIN KINASE C ACTIVITY AND PROTEIN PHOSPHORYLATION IN MOUSE SPERM

マウス精子において, 透明層により誘起される先体反応過程でカルシウム, リン脂質依存性プロテインキナーゼ (PKC) の関与が考えられている. 本研究にて, マウス精子細胞質画分に PKC 活性が検出され, PKC を活性化するホルボルエステルである TPA の処理により, PKC 活性ほ膜画分に移行することが認められた. 次に, マウス精子を [^<32>P] で標識し, 2次元電気泳動で精子蛋白質のリン酸化反応を解析した. TPA あるいはカルシウムイオノフォア A23187 処理により特異的な変化はみられなかったが, A23187で処理した後, さらに TPA を添加することにより, 215kDa と 35kDa の蛋白質がリン酸化された. 精子へのカルシウムの取り込みをみてみると, TPA と A23187 の存在下でそれぞれ単独の場合と比較して約2倍のカルシウムの取り込みがみられ, 相乗効果が示唆された. さらに, ヒト膵臓癌由来の精製 PKC を用い, in vitro においてマウス精子細胞質画分に含まれる蛋白質のリン酸化反応をみると, 215kDa と 35kDa の蛋白質が in vivo と同様にリン酸化され, これらの蛋白質は PKC の基質であることが証明された. 以上の結果より, マウス精子に PKC 活性と PKC 依存性蛋白質リン酸化反応が証明され, 先体反応における PKC の関与が示唆された.
