DIRECT INHIBITORY OVARIAN EFFECTS OF PROLACTIN IN THE PROCESS OF OVULATION

Prolactin (PRL) の卵成熟と卵胞破裂機序に及ぽす直接作用をin vitroでの家兎卵巣灌流装置を用い検討した。成熟日本白色家兎を用い, (1) 一側卵巣を灌流液のみ (対照群), 対側卵巣をporcine PRL 10^2, 10^3 ng/ml (実験群) で灌流し, in vitroにおける排卵, 卵成熟, steroid生合成に及ぼす影響について検討した。(2) 一側卵巣を各種濃度のPRL (10^3, 10^2, 10 ng/ml), 他側卵巣をmedium 199のみにて灌流, 30分後に両側卵巣にhCG (50 IU) を投与し, hCGによってin vitroで惹起される排卵に及ぼすPRLの直接作用について検討した。(1) PRL単独投与は, in vitroで排卵を惹起せず, 灌流12時間後の卵胞卵の80%以上はgerminal vesicle期であり, PRLは卵成熟効果を有していなかった。PRL群におけるprogesterone (P_4) 生合成量は6.1±0.6 ng/mlであり, 対照群との間に有意差は認められなかった。(2) PRL (10^3 ng/ml) はhCGによってin vitroで惹起される排卵を有意に抑制した (ovulatory emciency: PRL 25.8%, 対照群76.2%)。PRL濃度と排卵抑制効果との間には用量反応性が認められた。しかしhCGによって惹起される排卵卵子, 卵胞卵の成熟度に影響を与えなかった。PRL添加群のP_4生合成量85.2±11.7 ng/mlと, 対照群111.8±25.7 ng/mlとの間に有意差は認められなかった。しかしPRL (10^3 ng/ml) は卵巣におけるestradiol生合成を有意に抑制した。PRLは卵核の成熟になんら影響を与えないが, 排卵, とくに卵胞破裂機構に抑制的に作用する。しかしPRLの直接作用は, P_4産生を介していないことを確認した。
