INVOLVEMENT OF PLASMINOGEN ACTIVATOR ACTIVITY IN THE PROCESS OF OVULATION

卵巣内の線溶系酵素と排卵との関連が注目されているが, 今回排卵過程時に卵胞破裂機序におけるtissue-plasminogen activator (t-PA)活性の意義について検討を加えた. 家兎をhCG (100 IU)静注にて排卵誘発し, 排卵過程における成熟卵胞内のt-PA活性の変動を検討した. 測定法としては, 卵胞の2M KSCN-HC1抽出液にstimulatorとしての可溶性フィブリンを加え, 活性化されたt-PAが添加plasminogenをplasminに変換する際に合成基質から遊離される発色基質を吸光度(405nm)で測定した. 卵胞内t-PA活性は未刺激卵巣では1.54±0.08IU/g tissueであったが, hCG投与1hr後より増加し, 4hrで31.9±5.6IU/gの頂値に達し, その後低下した. 8hrで再び増加(27.5±5.8IU/g)し, 10時間で前値に復した. このように成熟卵胞内のt-PA活性は, 排卵過程で二峰性の変動を示した. in vitro の家兎卵巣灌流系においてhCG (50 IU)を投与し, 排卵に至るまでの卵胞内t-PA活性を測定した. また排卵過程における成熟卵胞および未熟卵胞のt-PA活性を比較検討した. in vitroの系においても80%前後のovulatory efficiencyを有する成熟卵胞内のt-PA活性は, hCG投与4hr後に28.4±4.2IU/gの頂値に達し, その後漸減した. ovulatory efficiency 20%以下である未熟卵胞のt-PA活性は, hCG 投与にもかかわらず12.4±1.9IU/gであり, 成熟卵胞との間に有意差(p<0.01)が認められた. 排卵過程における成熟卵胞内のt-PA活性は, 二峰性の変動を示し, 未熟卵胞においては明確なt-PA 活性の変化が認められなかった. 成熟卵胞におけるt-PA活性の変化は, 卵胞破裂機構に重要な役割を果たしていることが示唆された.
