Multiplication of Pinellia ternata by Callus Culture of Leaf Segment

Callus culture of Pinellia ternata has been established in which the leaf obtained from the tissue culture of tuber segment was inoculated onto Murashige and Skoog's (MS) medium supplemented with 2 ppm 2,4-dichlorophenoxyacetic acid (2,4-D) and 1-2 ppm kinetin. Regeneration from callus segment was stimulated in a medium containing 1 ppm a-naphthaleneacetic acid (NAA) to produce 50.7 plantlets per callus segment. The callus segment could be sub-cultured to multiply callus retaining its regeneration potential. As regards the effects of low temperature storage at 4℃ and 0℃ on callus growth rate and regeneration capacity, the storage of callus segment at 4℃ was found to increase the propagation capacity of callus and to its regeneration capacity compared to the control. When the callus segment was stored at 0℃, the regeneration potential and the multiplication capacity of callus decreased and finally disappeared after 20 weeks.
