Bioactive Constituents of a Human Placenta Hydrolysate : From Biogenic Stimulants Reported by Filatow to Biological Markers of Collagen Generation

Not only animal and human tissues, but also plant materials under stressed conditions have been widely used as original sources of the so-called "biogenic stimulant" reported by Filatow since 1933. Among plant sources, the fraction obtained from Aloe (A. arborescens) kept in cold storage at 4-5℃ under the condition of darkness for 12 d, was used as a biogenic stimulant and showed relatively higher levels of malic and tartaric acids than the controls. Increased urease activity was also reported. We examined the effects of Aloe arborescens var. natalensis (Kidachi aloe in Japanese), stored under cold and dark conditions for 10 d, on the phagocytosis and NBT-reduction of neutrophils from adult bronchial asthmatic patients, and found significant stimulation of anti-inflammatory activity due to phagocytic and NBT-reductive activities in the stressed materials. Afzal et al. analyzed our study on the biogenic stimulant of Aloe arborescens var. natalensis as follows: The stored cold and dark conditions were just right for the hydrolysis ofphospholipids releasing arachidonic acid to synthesize prostanolids involving endogeneously present cyclooxygenase in Aloe vera gel. In placenta stored at 2--4℃ for 5 d, nitrogen protein and glucose contents decreased by half, and lactic acid and non-nitrogen protein contents increased by 150%. These findings indicate that not putrefaction, but autolysis, glycolysis and lactic acid fermentation proceeded gradually under the conditions employed. In the present study, we isolated cyclo(trans-L-Hyp-L-Ser) together with δ-Hyl from the hydrolysate of human placenta (Laennec), and examined the biological and pharmacological activities of Laennec. Our results show that both compounds are closely linked to collagen generation in the placenta.
