Simultaneous determination of ten bioactive constituents in Eucommia ulmoides leaves and Tochu tea products by high-performance liquid chromatography-diode array detector-mass spectrometry (HPLC-DAD-MS)

杜仲の葉を原料とする杜仲茶は日本,中国,韓国で健康嗜好品として広く消費されている。そのため,これら製品の品質管理は安全性や有効性の評価のために重要である。高速液体クロマトグラフィー・ダイオードアレイ検出器/イオン噴霧型イオン化質量分析計(HPLC-DAD/ESI-MS)を使った方法により杜仲葉,杜仲茶製品中に含まれる10種の活性成分の同時定量法を確立した。これらの成分には2種のイリドイド,2種のフェノール類,6種のフラボノイド類が含まれる。ピークは杜仲葉や杜仲茶製品から単離した標準化合物との保持時間,オンラインでのイオン噴霧型イオン化マス(ESI-MS)スペクトル,紫外線吸収データを比較して同定した。10種の化合物の検量曲線は定量範囲内で全て良い直線性を示し,回帰率はR>0.9994であった。この方法は精密でかつ精度も高く,日内変動,隔日変動はそれぞれ1.8-3.0%,3.5-5.9%であった。また全体の回収率は95.4-106.1%であり,その相対的標準偏差(RSD)は分析した10種の化合物で3.6%(n=3)であった。この非常に選択性があり,すぐれた感度を持つHPLC-DAD/ESI-MSによる分析法によって杜仲茶製品や杜仲葉の成分に関する幅広い化学的情報を得ることができる。
