In Vitro Translation of α-Amylase mRNA from Dry Wheat Embryos

The poly(A)^+RNA fraction was separated from ungerminated wheat embryos and translated in both wheat germ extract and rabbit reticulocyie lysate systems. The polypeptides synthesized in vitro were immunoprecipitated using rabbit anti-wheat a-amylase antibody. Fluorography after SDS-polyacrylamide gel electrophoresis showed a band of molecular weight of 43,000, which was the same as that of the a-amylase precursor polypeptide. The a-amylase precursor syn-thesized from the mRNA of dry embryos belonged to the group II isozyme subset, characterized by an acidic isoelectric point.
